Yes! As long as you have enough actions to spend, you can Interpose and Defend at the same time. However, you won't be able to do either reaction again until your next turn ends since each is limited to 1/round.
Can you Interpose against an area-of-effect attack?

If one can be pushed out of the affected area, AoE attacks can be Interposed.
How does Interpose work with conditional attacks?

If it makes sense that the hero could push their ally out of the way of the attack (e.g., the ally isn’t Grappled), it works as normal—the Interposing hero suffers the damage and other effects as normal.
